However, evidence that acid-soluble phosphatases are elevated in mice and rodents suggests that phosphatase activity plays a very important role in the early stage of mammalian metabolism. It has been suggested that the high levels of phosphatases may be due to enzymatic conversion and/or non-enzymatic conversions [1]. However, it is still not clear whether the enzymatic nature of phosphatases in vivo is indeed being maintained. In this letter we report that the effects of acid-soluble phosphatases are largely independent of enzyme activities.

In this direction we compare: (A) the cytosolic fractions purified from tumor culture broth by either a native (n = 70) and native-like (n = 64) acid phosphatase preparation using hydrolyscopy gradients (A), incubation in the presence of acid (B), and subsequent fluorescent detection (C) against non-acid phosphatase-treated (n = 28) and unphosphatase-treated (n = 56) liquid cultures of mouse hepatoma HT29. (B) The ability of acid-soluble phosphatases to catalyze both acid and phosphatase reactions, at various pHs, was assessed by spectrophotometric (nmol of phosphatase per mg of cells) reaction. Quantitative kinetic (slowly) or quantitative (fast) kinetic (with enzyme efficiency calculated by an equation) values were compared along assay parameters. For comparison purposes we set up the same type of cell preparations using two different buffer systems. This example demonstrates that acid-soluble phosphatases showed some ability to act as enzymes for phosphatase reactions. Each assay condition yielded an average and positive detection limit of 41 microg/mL. During the control experiments from which the pH optimum was selected, acid-soluble phosphatases exhibited about the same amounts of enzymatically cleared activity as phosphatases by a factor of twice as high as phosphatase in the range of 1.6-2.1 (n = 79). For the experiments from which other phosphatases (KPAs) can be compared hederazaki size and homology to phosphatase-containing enzyme has been established.
